Scored for wind instruments and percussion and about 10 minutes in length, this work was premiered on March 29, 1965 by the Los Angeles Neophonic Orchestra under the direction of Stan Kenton. The same group made a studio recording of the piece shortly thereafter. Reviewing the concert in the Los Angeles Times, Mimi Clar was not entirely positive about the program as a whole, but wrote, "there was some worthwhile music, most particularly a Prelude and Fugue by John Williams, based on the brooding chromaticisms of a melodic tone row."

In the liner notes for that LP, Noel Wedder wrote:

A medievally-inspired introductory line begins softly, then swells and gathers momentum as brass and percussion are called upon to deliver a more formidable message. The variety of ideas subtly but precisely woven through the score perfectly complement Williams' geometric, liturgical-in-mood constructions. Thematic lines, forcefully etched against shifting counter lines, heighten the suspense and excitement as the Orchestra keeps surrendering one aural experience after another. Dedicated to pianist Claude Thornhill, whom Williams knew as a child, Prelude and Fugue is an outstanding example of the awesome beauty and incredible power that can be unleashed by the Neophonic Orchestra.
